1. Thaw samples at room temperature. 2. Weigh 10 g fresh silage, transfer to glass jar blender. 3. Add 100 mL Milli-Q water to blender. 4. Blend for 30s at intermediate rate. 5. Filter mixture using PYREX Gooch Type Filtering Crucible and vacuum filtration equipment, keep liquid filtrate. 6. Transfer 2 mL filtrate to centrifugation tubes, add 2 mL extraction solution. 7. Vortex for 30s at intermediate rate. 8. Centrifuge at 10,000 x g for 15 min at 4 ºC. 9. Recover liquid phase using a plastic syringe, filter through 0.45 μm PES membrane, collect 500 μL filtrate in chromatographic vial. 10. Prepare calibration curves for lactic acid and SCFAs. 11. Analyze calibration curves using RP-HPLC-DAD. 12. Identify and integrate chromatographic peaks. 13. Place 500 μL extract in chromatographic vial, analyze using RP-HPLC-DAD. 14. Identify and integrate chromatographic peaks. 15. Set chromatographic conditions: column temperature 30 ºC, injection volume 20 μL, Hypersil GOLD C18 column with guard column, working wavelength 210 nm, gradient mode with 0.750 mL/min flow rate.
