Abstract: Highlights•We propose a new method for high-throughput screening of CTA and validate its effectiveness.•We identified serine protease PRSS41 as a novel cancer-testis antigen that can serve as a target for immunotherapy of gastrointestinal cancer.•PRSS41 can serve as a diagnostic biomarker that is frequently overexpressed in gastrointestinal cancer due to promoter DNA hypomethylation.•Promoter DNA methylation level of PRSS41 (cg08104780) predicts poor prognosis in gastric cancer.
